Annex D: List of Canadian microbiological screening and cultural methods for Salmonella, Listeria monocytogenes, Escherichia coli O157:H5 recognized as equivalent by the FSIS

As of October 17, 2016 the CFIA currently has equivalency with the United States – FSIS for the following screening and cultural methods for E. coli O157:H7, Salmonella spp. and Listeria monocytogenes.

For E. coli O157 and Salmonella testing in meat products, a 325g portion must be analysed.

For Listeria testing in RTE meats and egg products, a 125g portion must be analysed. Swabs of environmental surfaces may be tested individually or composited (up to 10 swabs).

For Salmonella testing in egg products, a 250g portion must be tested to meet domestic requirements. USDA/FSIS requirements for egg products exported to the U.S. are for analysis of a 100 gram test portion.

Approved methods can be obtained from the Health Canada Compendium of Analytical Methods site. Note that the most recent published version of the method should be used. The "application" section of the method chosen must be appropriate for the intended purpose.

E. coli O157:H7 in Meat

  1. MFLP-16, Detection of E. coli O157:H7 in Foods – Assurance GDS™ for E. coli O157:H7 Gene Detection System, with the following stipulations:
    • incubated for 10 to 18 hours at 42°C
  2. MFLP-30, Detection of Escherichia coli O157:H7 in select foods using the BAX® System E. coli O157:H7 MP
  3. MFLP-12, Identification of E. coli O157:H7 and Verotoxin-producing E. coli O157 Non Motile (NM) by the Adiafood Rapid Pathogen Detection System, a Real-time PCR technique
  4. MFHPB-10, Isolation of Escherichia coli O157:H7/NM from foods and environmental surface samples, with the following stipulations:
    • typical E. coli O157:H7 colonies on selective agar that do not confirm as a toxin producer must be further characterised by PCR testing of toxin genes and H7 genes. If the lab does not have this capability, isolates must be submitted to the CFIA for this testing.
  5. MFLP-76, The DuPont Qualicon BAX® System real-time method for the detection of E. coli O157:H7 in raw beef trim and raw ground beef
  6. MFLP- 98, Detection of E. coli O157:H7 in food products by the VIDAS® UP E. coli O157 (including H7) method
  7. MFLP-87, Detection of Enterohemorrhagic Escherichia coli O157 (EHEC) in Food Products and Food Ingredients by the VIP® Gold for EHEC Method

Salmonella spp. in Meat and Eggs

  1. MFLP-29, The Qualicon BAX® System Method for the detection of Salmonella in a variety of food and environmental samples
  2. MFLP-20, The Genequence® Salmonella Microwell Assay for the Detection of Salmonella in a Variety of Foods
  3. MFHPB-24, Detection of Salmonella spp. in Foods by the VIDAS SLM™ Method
  4. MFHPB-20, Isolation and Identification of Salmonella from Foods and Environmental samples

Salmonella spp. in Meat

  1. MFLP-32, Identification of Salmonella species using the Adiafood Rapid Pathogen Detection System, a Real-Time PCR Technique

Listeria monocytogenes in Meat, Eggs and Environmental samples

  1. MFLP-28, The Qualicon Bax® System method for the detection of Listeria monocytogenes in a variety of food
  2. MFHPB-30, Isolation of Listeria monocytogenes and other Listeria spp. from foods and environmental samples

Listeria monocytogenes in Meat and Environmental samples

  1. MFLP-77, Detection of Listeria monocytogenes and other Listeria spp. in food products and environmental samples by the VIDAS® Listeria species Xpress (LSX) method

Listeria monocytogenes in Environmental samples

  1. MFLP-15, The detection of Listeria species from environmental surfaces using the Dupont Qualicon BAX® System method and direct plating
  2. MFLP-05, Detection of Listeria spp. from environmental surfaces using the 3M™ Molecular Detection System Test Kit

In addition, FSIS will accept all methods for the above pathogens published in the USDA/FSIS Microbiology Laboratory Guidebook.
